Alright, so I'm getting money from some relatives to put into my checking account, which I'll use to make a large initial deposit on the room of my choice. The thing is, I don't know which room to play on. I've narrowed it down to Paradise for the variety of gametypes they have, Full Tilt for the practice against skilled players, and PokerStars because this is what all my friends from StarCraft played, so this is what I've been familiar with.
I haven't really played a ton online, aside from a few play money sitngos and the Freeroll tournaments on PokerStars. Other than that I just have a few home games I have with different groups of friends. So should I put all my money into one room to build up a bankroll first (and if so, which room), or should I put a minimum deposit in each room and test them out? I've read here that Full Tilt is really hard to build up a roll, but it has the best overall skill level of players, and from what I've seen trying out Paradise, it's got plenty of newbies (at least as many as PokerStars).

I vote PokerStars, 2nd largest site with largest selection of small games for new (smaller) bankrolls.

Azure-
Get a Neteller account and load your bankroll into it to get started!! (verify your account so the deposit is FREE...use EFT, and NOT insta-cash).
Stars is a good choice for the sheer variety of games, but a bad choice in that they offer no signup bonus.
I vote for Paradise. You will get bonus-money just for signing up, which will be added to your BR in small bits as you play. If you decide you don't like Paradise, then you can withdrawal your money to go play somewhere else...the bonus will still be there if you ever want to come back. Paradise has NL$2 to get your feet wet. As you're comfortable, move up.
Keep BR101 in mind, though...unless/until your BR is $500+, you shouldn't play NL$25. 20 buy-ins is the rule of thumb.
If your BR is $500+ and you're comfortable playing NL$25, you have a LOT more options than the three sites you named.

of the ones you mentioned I say Pokerstars. but there is no reason to limit yourself to just one poker site. I have accounts on about 10 right now. Get a neteller account and start doing some bonus whoring. Some other sites I'd recommend are Partypoker, Bodog, and Absolute Poker.

Absolutely.
Azure
Party's smallest tables are NL$25 so you'll need $500 to play there...but they have the best and easiest bonuses!
Absolute Poker's enticing bonus doesn't clear well at >NL$100. Unless you're planning to start your BR off at $2,000, this is NOT the site for you. They OFFER micro tables, but you won't clear a bonus playing them. Absolute software = the suck, too.
My vote stays: Paradise Poker. Microtables and a bonus that clears OK at NL$25 but will clear (albeit slowly) at NL$10.
You definitely don't need to stick to just one site. Neteller will help you move your money for bonus after bonus after bonus. I opened 6 new accounts this month to chase new bonuses.
